I just got a job putting down a 2×6 SYP T&G floor in a timber frame house. It will be used as the ceiling in the basement as well as the finished floor on the first level. The timber frame is 30×40. Should I start in the center or at one wall? What type of fastener should I use? I am planning on using adhesive on the floor joists as well.

I start at one end and work across, using 8d ring shank in the tongues.
You will need some bar clamps and some wedges and screws also. Maybe some 12d for the boards that want to fight with you

I'd do a quick layout to see how it looks best.. you don't want one wall to have a sliver or something just to get it to fit. Piffin is right about fighting with some boards.. they may look straight in the pile but as you are putting them down they seem to love to fight you.
I'd also check the moisture of the boards.. Kiln dried doesn't really mean what you think it does so the boards could shrink a lot over the first heating season and even possibly leave gaps. Then you really have a battle on your hands..
Careful about the adhesive. I thought I was extremely carefull and yet some boards oozed it out and I didn't catch it untill it was too late.
I used rink shanks in mine Stainless steel since they were going into white oak.. My first boards I screwed down and that was a real wrestling match. I eventually predrilled all of them and even then if I was a little overly zealous in tightening them the heads would snap off..
