how far from an a/c supply duct must the return be in a room serviced by both supply and return.

As far as possible.
There are no code issues that I'm aware of for supply and return proximity to each other. Just common sense.
I assume your referring to a great room or living room. No return air in the kitchen(and a few other common sense places). Return air duct needs to be a min. of 10' away from a fireplace or any other vented appliance.
For AC, it should not be on the floor.
I once heard a presentation by a mechanical specialist who said the location of the return makes little difference.
He held up a match about 1 foot from his face and blew it out. He lit another match about 1 foot from his face and then started sucking.
he said "no matter how hard I suck, I can not suck that flame out"
